I did my entire registry online, but for the bigger stuff you might want to actually go into the store and look/test it out first (swing, pack n play...)
I browsed target.com for alot of stuff and made notes so I wouldn't forget once I got there and I did register for a few things before, so I would say it really just depends on what you prefer.
It's easier to start it online because then you can just go to the store and grab a gun. You don't have to sit there with someone to give your info and start out your registry.
It's also great to test stuff in the store, add it, but then go back and read reviews of everything that you picked. I also had my mom (owns a daycare) and my sister (mom of 2) to look at my registry to make sure I had all the right stuff on there
DH and I went to the store first (Buy Buy Baby and Babies R Us) because our top priority was the 'gear', and we wanted somebody knowledgeable there to answer questions, etc. (though I definitely recommend doing your own research before you go in). I definitely would recommend it- it was nice to start off by going section to section, seeing what was available and what the various options were..And if you have Buy Buy Baby, I recommend going there first -they are SO informative, without pushing you toward a more expensive item. Truly helpful.
The main things I registered for online were simple items like onesies, towels, washcloths, etc... and the highchair, because the model I wanted was not in stock at the store.
Also keep in mind, you can always go back home and change stuff! I did a lot of shuffling for a week after the registry was 'finished'
Some items are available online only...other then that the benefit to going into the store is you can browse the aisle and see things you might not have thought about.

If you start the registry online you are still eligible to pick up the packet of free stuff during your next visit to the store.
Do you still get a completion coupon? I would much rather just register at home since I'm just making a list of things to get a coupon for and know pretty much what I'll put on it.
I found that going to the store to see what they offer at the store is a good idea because when you register for some things on line you have to check and make sure they offer it in the store. Many older people do not like to buy online and have it shipped. Also many people buy last minute and there would be no time for the gift to be shipped espically from Target, it seems they are always slow on the shipping, at least in my experience.
