I usually test once a day until I notice my OPK getting a little darker. Once I notice that I start testing twice a day. I'm still learning my cycle so next cycle I may start testing a bit later.
Yay for AF! I use the CBEFM so only POAS 1 x per day. However, it has missed my surge every month, so maybe testing multiple times per day is beneficial. I know I did O because I got pregnant using the monitor. Hopefully it works this month!

I POAS twice a day (late morning, early evening). I start about CD 7 which is when I usually start to see wet CM. I tend to O a bit early, between days CD10-12. If you are just starting to use them there is an opk test chart on pee on a stick.com that tells you what day to start testing on. That website is full of great info!
