We've got a cool aquarium and the Tampa Theater has a summer movie series where they show old movies. It's the coolest movie theater ever. Check out their website. For night life there are fun clubs and restaurants at Channelside. They have a website too. I haven't stayed at a lot of hotels in Tampa (obviously since I live here), but the Marriot Renaissance that's attached to the International Plaza mall is really nice. Lots of shopping at the mall and the concierge can get you a table at the Cheesecake Factory with no wait.
If you're into theme parks, Busch Gardens is pretty cool - they have a kids section, but I tend to think of it as more adult because of all the roller coasters. If you're traveling with kids who are strong swimmers, Adventure Island is a fun water park. Pass-a-Grille beach is nearer St. Pete, but it's gorgeous - the sand is super soft and relaxing. You can check out a Rays game at Tropicana Field in St. Pete if they're in town, or shop at International Plaza. I'll second the Aquarium, too... very cool, if you're into that.

For a romantic evening, after dinner go to Bern's Steakhouse in South Tampa for dessert in the dessert room. You can expect to pay $10 for dessert and $4 for a coffee, but SO DELICIOUS and a very very romantic setting. They have a HUGE dessert wine and liquor list also. Get the macadamia nut ice cream - made there and just so good.
I would strongly suggest that you stay at Tampa Marriott Waterside Hotel & Marina~ It's downtown and offers lots of things to do. All of the things that the other girls mentioned are close by too!
