DD can go through up to six diapers in a night but she only really wants to eat twice. So I started packing her up at night with a large bamboo flat folded orgami and a O/S Assunta cover. Its softer/stretchier than the thirsties duos. Now she can do two diapers that way, one overnight and one around 7am. I get to sleep till 9am after that 7am diaper change.
The only problem is that the diaper reeks in the AM, especially the first one. Like a strong urine smell. The cover also smells and is damp.
I want a wool cover for over-night but could only afford one. However I sew and could make wool covers from old wool sweaters if I can find some at the thrift stores this late in spring. But what would I do to make my homemade covers usable? I only do laundry once a week. Would I need to make six covers or can I reuse a cover more than once by washing it out in the sink and line dry every morning? What are my alternatives?

First, your current ON solution the diaper smells because as urine hits air it turns into ammonia. So the pee that LO does early in the night is likely turning to ammonia during the night and causing the smell in the morning. Nothing to be concerned about other than you might want to soak and/or rinse that diaper and cover out before putting in your wetbag to ensure that it gets all the way clean once you do your laundry routine on it.
I'll caveat this by saying that I have not use wool yet. Like you I am hoping to find time to upcycle a thrift store wool sweater or two this summer. But from my research:
Wool has anti-mircobial properties
If you lanolize it "right" it shouldn't leak, but instead will sort of "absorb" the urine that gets on it. Search YouTube for info on how to lanolize "right", and from reading on this board, you want to use a little more than the small pea sized drop of lanolin that they say to use.
You only need to wash every week or two rather than after every use. Therefore you really only need 2-3 covers.
I rinse my covers between uses and hang dry them unless they get pooped on then I rinse 'em and chuck 'em in a wet bag separate from the wetbag that I put my prefolds and cloth wipes in. Once a week I wash everything that was used even slightly.
This isn't totally true. You can definitely machine wash interlock.
