The Kindle isn't backlit and is an ink that like an actual book. The battery will last longer, quite a bit longer. You can get a model that has 3G on it and you don't pay any monthly fees for it or anything. Great return policy with Amazon. You can get a gift card for Amazon and buy books with it.
The Nook Color is back lit. You have to connect to the internet via WiFi or your USB internet connected computer to put books on it. You can root it and it's an Android Tablet, but cheaper than buying an actual Android Tablet. You can read books on it from Amazon if you root it. There is a glare on the screen because it's a normal screen in sunlight or certain conditions indoors. You can get books in color, such as kids books or comics! It's fun for kids because of that aspect. Your book covers are in color as well. There are kids books that are "read to me" and that's neat.
I have both and there are pros and cons to each. I'm giving my H my Nook color so he can use it as a tablet and I can use the Kindle as a regular e reader.

I have a nook color and love it, best gift I have ever been given. You can get magazine subscriptions on it too. An e reader is the actual device, the nook or kindle.
