Try giving it every feeding. Our pedi told me that Mylicon isn't systemic (doesn't enter the blood stream). Instead it reacts directly with the milk. So, if it doesn't go in with the milk, it won't work.
We HAVE to give it to our boy at every feeding or we pay for it with bad gas that is painful for him. Mylicon makes it manageable!
