Dinosaur Land somewhere in VA. Or Jekyll Island. DH says it rocks, but I've never been. Savannah, GA for the history. NYC, NY. Went there once, loved it, really wanna go back.
Intercourse, PA. Because I guarantee you won't be able to get out of the town without giggling at the name. Plus you can get one of the fun bumper stickers and other merchandise that says "I Intercourse (PA)."
And then you can drive north and visit me.
Why not Blue Ball too in case you don't quite make it to Intercourse

Also, I'm from the NYC-area, and I guess Manhattan is a must-see if you're driving through. There is a ton of kid-friendly crap in Pennsylvania. Sesame Place, the Please Touch Museum, the Franklin Institute Science Museum (maybe for older kids; I loved that shiit as a girl scout).
This is probably out of the way, but Lake Placid in upstate NY is one of my favorites. They have all the former Olympic stuff and an awesome town for shopping/drinking/eating. And the Adirondacks are beautiful.

NYC (venture out to the other suburbs for what I consider a truer NYC experience I'd recommend Brooklyn if you only have time for 1 other suburb.
One of the many kid friendly attractions in PA - Sesame Place, Hershey Park, Crayola Factory, Dutch Wonderland, Great Wolf Lodge, Strassburg Railroad, Please Touch Museum, Franklin Institute, Longwood Gardens, Morris Arboretum.
I agree with Acadia National Park. It is absolutely beautiful, especially in the fall. We used to go hiking every October before the boys came along. Ogunquit is also amazing!

But if you are going east as far as Maine, mayswell drive the 6-or-so hours and come hang in PEI, Canada. I know I'm biased, but this place is farking GORGEOUS in the summer.
