have someone sit with him if possible. Also plan to make a few quick stops - kept DS from feeling trapped in his seat and gave him a chance to stretch his legs.
We've done several >3hour drives with DS. The roughest was the first one after we switched carseats, but a neck pillow helped that. It really depends on how your LO usually does in the car. DS does pretty good usually. I def use a mirror, so I can see him. This is especially i,portant to me when I am traveling alone with him. I usually leave when it's time for him to take a nap. Then he sleeps the first part of the trip. We'll stop once on the way to nurse and diaper change, and then he either sleeps, looks out the window or plays with a toy for the rest of the trip. The toys I give him are his teething beads and his elephant, which is like floppy with a rattle head, celophane stuffed belly and chewable feet.
ETA: We haven't sat in the back with DS since he was like 2 weeks old.

We drove 13 hours down to florida and DD did really well! She slept a lot but when she wasn't sleeping I had toys that hung from her carseat that she played with plus a few extra toys that she had in her carseat with her.

I also packed a "goody bag" for him with books, toys, plastic water bottle, etc... things that he hadn't seen/played with in a long time (diff. textures, colors, sounds etc) when he'd get fussy, I handed him a new toy and it would keep him interested for a long time! I also kept puffs/sippy cup out and he snacked on that for a while too..

Some of This.
Plus, we make sure we have a bottle or 2 ready that way she doesn't have to wait long due to us going over a Washington mountain Pass. Now, that Lo is getting bigger we may also make sure snacks are on hand since she is eating some solids.
We don't have to take out LO as long as the diaper is dry and she is asleep. But we have been fortunate to plan around naps also pr bed time. Plus, the trips we have taken so far LO did while I was pregnant not sure if that is cheating on being used to it or not.

Me, dh, SIL, a friend and possibly one of SILs friends will be in the back with him (yay suburban) these tips are awesome ladies!
Most of my long trips with DS have been alone. Since you are going to have so many people, maybe it wouldn't be a good idea to wait for a nap time, because he might have trouble sleeping with so many people in the car. Maybe leave just after a nap?

I sat in back with DS (helped a ton!)
I also packed a "goody bag" for him with books, toys, plastic water bottle, etc... things that he hadn't seen/played with in a long time (diff. textures, colors, sounds etc) when he'd get fussy, I handed him a new toy and it would keep him interested for a long time! we played "peek a boo" sang songs, etc... GL!
