You should ask about hours, late pick ups, extra fees (i.e. in the summer we are charged an activity fee - its only $50 for the whole summer, but still - it was unexpected) also what things you ahve to provide vs. daycare, what their cleaning schedule is, also what education everybody has - not just the director, but the actual in the classroom teachers, how long everyone has been there (i.e. turnover). When you find a good daycare you will know it/feel it - its always a little sad to leave your LO, but you should feel like you are leaving your LO in a loving place w/ people who will care for him/her.
Ditto Kathryn. Also ask about the sick policy. If they call you at work, do they expect you there within the hour, is there some flexibility, etc.? Do they mind you calling to check up during the day? What is a typical day like? Ask Kathryn said, you'll know the daycare that's right for you.

oh - yeah - ask about "curriculm" - DD's daycare has a week curriculm that they start around 6 months. Its not like an academic curriculm, but there is a monthly theme and they reinforce it w/ the songs they sing, books they read, they have specific activities that they do - i.e. exploring different textures. Now, in the toddler room, they have more weekly curriculum. None of its hard and fast, but once your LO is past the "lump" stage, you want to make sure they are providing opportunities for learning.
I would suggest after your LO arrives and as it gets closer to your start date go spend some time with your LO at the DC. I had no idea my 1st choice had a change in teachers in the infant room. I left there in tears my gut was not happy with this DC any longer. I had kept my name on the list at my second choice I went there with LO and left there feeling great. Like Kathryn said it is a feeling you will know which is right for you.
